Duwaynw Burnside was born in late sixties in Senatobia, in Mississippi. He grew up listening to his father R.L. Burnside, and family and neighbors play music: guitar driven Mississippi hill country blues. He was the son of legendary blue singer, songwriter, and guitarist R.L. Burnside, who lived most of his life in and around Holly Springs, Mississippi. He played music for much of his life, but did not receive deserved attention until early 1990's. The musical influence was not restricted to his father and his family but neighbors and the radio played a vital role too. He picked up guitar even before he was old enough to hold himself.
While growing up, Duwayne use to play guitar behind his father, but more frequently for local club owner Junior Kimbrough and Soul Blues Boys. Since Memphis, Tennessee were not that far, he began playing and sitting in with seasoned and well known musicians that frequented town, such as Little Jimmy King, Bobby Blue Band and many others. Simultaneously he was playing in R.L. Burnside's family band Sound Groove, which was combination of hill country sound as soul and R&B that was splurging out of Memphis at that time. The credit for Duwayne's outstanding rhythmic style goes to the two new styles and his growing experiences of conventional style. This cultivated in him the ability to blend with all styles of music and any musician. While playing rhythm Duwayne also paid attention to all other areas of music and stepped up to fit in anywhere he was needed. He tried his hand at playing drums, bass or guitar. Soon Duwayne's leads began to sound fused with Albert King and the heavy soul feel that came from his experience of playing at early stage.

Duwayne did some recordings with his father and the Sound Groove on High Tone and Fat Possum Records. After that he moved to Memphis and opened his own club, Burnside Kitchen and Grill near Highway61. He was juggling between booking shows, cooking food, selling beer and use to play weekly, all at the same time. In mid 1998, Duwayne traveled to Los Angeles, California for the recording of his first full length record Live at the Mint as Duwayne Burnside and Mississippi Mafia Members inclided Cedric Burnside, Eddie Batos, Joe Hill from Alien Ant Farm, and David Kimbrough with Duwayne's father R.L. Sitting in on a few tracks. After coming back from his recording, Duwayne decided to put his bar businesses on halt and shift back home to Holly Springs and settle down for a while. In 2001, Duwayne performed with the North Mississippi All Stars for the first time in Birmingham, AL. He played rhythms and his leads with such perfection that it left everyone surprised, as it was his first show with the band. This success marked the beginning of his touring nearly for 259 days a year with the band and recording with All Stars on their third record Polaris and was also featured on two E.P.'s. In the dawn of 2004, Duwayne felt the need to pull reins and settle down back home for a while opening another club, Burnside Blues Cafe. He also formed a new band with old friends. The union is a heavy continuation of hill country and soul blues fusion. In 2006, Duwayne Burnside got nominated for 2006 Blues Music Award in the category of Best New Artist Debut- Under Pressure.
